摘    要:
目的观察Tween-80合并温热对人膀胱癌阿霉素耐药细胞株的逆转作用及其机制。方法采用MTT法进行体外药物逆转实验,免疫细胞化学染色显示处理前后耐药细胞P-糖蛋白表达情况。结果①Tween-80合并41℃温热作用前后,EJ/ADM对抗癌药物阿霉素、丝裂霉素、5-氟尿嘧啶等药物的敏感性显著增加(P<0.01);②Tween-80合并41℃温热能够协同抑制p-gp蛋白的表达。结论Tween-80合并41℃温热对人膀胱癌阿霉素耐药细胞的耐药性有逆转作用,逆转机制与其抑制p-gp蛋白的表达有关。

Reversal of drug resistance in the Adriamycin-resistant human bladder cancer cell lines EJ/ADM by Tween-80 in combination with hyperthermia, and its mechanism
Abstract:
Objective To investigate the reversal of drug resistance in the ADM-resistant human bladder cancer cell lines EJ/ADM by Tween-80 in combination with hyperthermia,and its mechanism.Method The reversing drug resistance was studied by MTT and the expression of P-glycoprotein were examined by immunocytochemistry.Result MTT cytotoxic assay revealed that the sensitivities of EJ/ADM cells to ADM,MMC,5-FU,HCPT,VCR were different according to presence or absence of Tween80 combined with hyperthermia(41℃)(P<0.01).The immuno-cytochemistry showed that the P-glycoprotein distributes in cellular membrane and cytoplasm,and after treated with Tween80 combined with hyperthermia(41℃),P-glycoprotein staining was redused in EJ/ADM cell.Conclusion The drug resistance of EJ/ADM cells could be reversed by Tween-80 in combination with hyperthermia(41℃).
