红芪总多糖体内抗肿瘤的实验研究

摘    要:目的:研究红芪总多糖(THPS)单用及与环磷酰胺(CTX)合用对荷瘤小鼠的抑瘤作用,并探讨其机制。方法:接种S180的昆明种小鼠腹腔注射低、中、高剂量THPS并联合使用CTX14天后,观察瘤重、抑瘤率、胸腺指数和脾指数,流式细胞仪检测T淋巴细胞亚群CD4 细胞和CD8 细胞数量。结果:与荷瘤对照组相比,中剂量THPS明显抑制小鼠S180瘤生长(P<0.01),增加胸腺指数(P<0.01)和脾指数(P<0.01),提高CD4 细胞数量(P<0.05);中剂量THPS与CTX合用后,与环磷酰胺组比较,明显降低瘤重(P<0.05),减轻CTX所致的胸腺指数(P<0.01)、脾指数(P<0.01)、CD4 细胞(P<0.01)和CD8 细胞(P<0.05)数量的下降。结论:中剂量的THPS具有抑制S180瘤生长,减轻CTX的免疫抑制作用,其机制与提高免疫器官的重量和T细胞介导的免疫应答有关。

Abstract:Objective:To investigate the suppressed e ects of total hedysarum polybotrys saccharide(THPS) alone and THPS combined with Cytoxan(CTX) on tumor-bearing mice and its mechanisms.Method:Kunming mice inoculated with S180 were given low,moderate and high dose THPS and THPS combined with CTX by intraperitoneal injection.Fourteen day later,tumor weight,inhibition rate,thymus index and spleen index were observed.CD4 and CD8 lymphocyte were counted though the ow cytometer.Results:Compared with the control,the moderate dose THPS showed signi cantly suppressed e ect on the growth of S180 tumor(P<0.01) and increased e ects on thymus index(P<0.01),spleen index(P<0.01) and CD4 lymphocyte count(P<0.05).Compared with the CTX group,THPS combined with CTX obviously reduced tumor weight(P<0.05) and delayed decrease of thymus index(P<0.01),spleen index(P<0.01) and the numbers of CD4 (P<0.01)and CD8 (P<0.05)lymphocyte caused by CTX.Conclusions:The moderate dose THPS showed signi cant suppressed e ect on tumor in S180-bearing mice and reduced immunosuppression caused by CTX through improving the weight of immune organs and T cellular mediated immune response.
