尼莫地平壳聚糖缓释微球制备工艺及性能研究

摘    要:目的研究以壳聚糖和阿拉伯胶为基质,制备尼莫地平缓释微球的工艺。方法以微球的药物包封率为制备工艺优化指标,得出成球的最佳制备工艺条件。结果最佳工艺条件为:搅拌速度400 r/min,pH 4.5,壳聚糖与阿拉伯胶重量浓度比为1∶1,戊二醛用量为1%。结论以最佳制备工艺条件制备载药微球,重现性好,工艺稳定,同时体外释放实验表明,该微球具有较好的缓释作用。

Abstract:Purpose To study the method which was used to prepare nimodipine microspheres with chitosan-Gum-arabic by means of a complex coacervation method in an emulsion system.Methods The orthogonal experimental design using the standard of drug encapsulation was applied to optimize the preparation procedure of the microspheres.Results The optimal preparation technique was as follows:400 r/min,pH 4.5,chitosan∶Gum-arabic=1∶1,and amount of glutaraldehyde is 1%.Conclusion This method was reproducible.The release of the microspheres in vitro indicated that these microspheres may prove better sustained release of drugs.
