饮食前后服氯氮平血药浓度对照研究

摘    要:目的:比较研究饮食前后服氯氮平的药代动力学和生物利用度。方法:采用交叉给药方案,19例精神分裂症病人空腹或餐后服用氯氮平,用HPLC测定血药浓度,并作副反应量表测定。结果:饭前服氯氮平达峰时间缩短,高峰浓度升高,生物利用度提高,两种服药方法间差异显性(P<0.05)。结论:饭前服药使较小的剂量达到治疗血药浓度,这样较快地控制精神症状,又减低了药物用量,可减少药物副反应。

Abstract:Objective To compare the pharmacokinetics and bioavailability of clozapine under conditions of fasting and after meal. Methods Clozapine was given to 19 schizophrenia patients under conditions of fasting and after meal in a crosover study, and serum clozapine concentration was detected by HPLC. Results Under fasting condition, the clozapine tmax was shortened, cmax was increased and the bioavailability of clozapine was raised. There were significant differences between two conditions (P<0.05). Conclusion Taking clozapine preprandially could lower the dosage of clozapine and control mental symptoms quickly; the side effects would be reduced.
