摘    要:
目的:探讨LPS在宫颈癌细胞株Hela细胞增殖和上皮间质转化中的作用。方法:体外培养Hela细胞,以LPS(10μg/ml)诱导后,利用MTT法检测细胞的增殖情况。qPCR检测上皮细胞标志物E-cadherin和间质细胞标志物N-cadherin、Vimentin的RNA水平上的表达。Western blot检测上皮细胞标志物E-cadherin和间质细胞标志物N-cadherin、Vimentin的蛋白水平上的表达。Transwell检测LPS对Hela细胞迁移能力的影响。结果:在LPS诱导后能够明显的促进Hela细胞的增殖。E-cadherin在RNA和蛋白水平上都明显下降,N-cadherin、Vimentin在RNA和蛋白水平上都明显升高;LPS诱导后,明显促进Hela细胞的迁移能力。结论:LPS能够诱导宫颈癌细胞株Hela细胞的增殖和上皮间质转化。

Abstract:
Objective:To investigate the effect of LPS on proliferation and epithelial-mesenchymal transition(EMT)of human cervical cancer.Methods:A549 cells cultured in vitro were treated by LPS(10μg/ml).The effect of LPS on proliferation was detected by MTT.The RNA level effect of LPS on epithelial markers(E-cadherin)and mesenchymal markers (N-cadherin,Vimentin) was detected by qPCR.The protein level of LPS on epithelial markers (E-cadherin)and mesenchymal markers (N-cadherin,Vimentin)was detected by Western blot.The effect of LPS on migration of Hela cell was detected by Transwell assay.Results:After treated by LPS,significantly promote the proliferation of Hela cell.The RNA and protein level of epithelial markers(E-cadherin)was decreased,and mesenchymal markers (N-cadherin,Vimentin)was increased.Transwell test showed that the migration potential of Hela cells was significantly increased after LPS treatment.Conclusion:LPS can induce cervical cancer cell line Hela proliferation and epithelial-mesenchymal transition.
