氟罗沙星片人体内外相关性的研究

摘    要:
 目的研究5个厂家生产的氟罗沙星片体外溶出速率与人体内生物利用度的相关性。方法应用RCZ-8A智能药物溶出仪测定不同厂家氟罗沙星片的溶出速率;12名男性健康志愿受试者po400mg氟罗沙星片,以尿药浓度法测定药动学参数,并考察体内外参数之间的线性关系。结果5种不同厂家氟罗沙星片的内累积排药量达到50%时的时间t与体外溶出t50二者之间呈明显的线性相关关系,回归方程为y=0.7853X+15.4946,r=0.9299。结论氟罗沙星片的体外溶出参数td与体内药动学参数tmax之间存在相关性。

Abstract:
OBJECTIVE To investigate the relationship between dissolution in vitro and absorption in vivo of fleroxacin tablets from five pharmaceutical corporations. METHODS Dissolution of fleroxacin tablets were tested with RCZ-8A dissolution tester. A single dose of 400 mg fleroxacin tablets was given to 12 healthy volunteers orally. Their pharmacokinetic parameters were obtained by using 3P97 program. RESULTS The correlation coefficient between the dissolution t_d in vitro and t_~max in vivo was 0.929 9.CONCLUSION There was a significant correlation between t_~d in vitro and t_~max in vivo.
