摘 要: | 目的观察阿魏酸哌嗪(Piperazine ferulate,PF)对单侧输尿管梗阻(Unilateral ureteral obstruction,UUO)模型大鼠肾小管间质的转化生长因子-β1(Transforming growth factor-β1,TGF-β1)、α-平滑肌肌动蛋白(α-smooth muscle actin,α—SMA)和Ⅰ型胶原(Collagen Ⅰ,ColⅠ)表达水平的影响,初步探讨其对小管间质纤维化的作用机制。方法将24只大鼠分为假手术(Sham—operated rats,SOR)组、UUO组、氯沙坦(Angiotensin Ⅱ receptor blocker,ARB)组和PF组。运用免疫组化技术半定量分析实验第14天各组大鼠肾组织中TGF-β1、α—SMA和ColⅠ的表达水平。结果免疫组化结果表明SOR组TGF-β1、α—SMA和ColⅠ均为少量表达,UUO组大鼠上述表达明显增加;ARB组和PF组TGF-β1、α—SMA和ColⅠ的表达均低于UUO组,但高于SOR组,两治疗组之间差异无统计学意义。结论阿魏酸哌嗪可以降低肾小管间质TGF-β1、α—SMA和ColⅠ的表达水平,延缓肾小管间质纤维化的进程。

Abstract: | Objective To explore the effect and mechanism of piperazine ferulate on regulating the expression of transforming growth factor-β1 (TGF-β1), α-smooth muscle actin (α-SMA) and collagen Ⅰ (ColⅠ) in renal tubulointerstitium in the rat model of unilateral ureteral obstruction (UUO) Methods Twenty-four SD rats were randomly divided into sham-operation group, UUO group, angiotensin Ⅱ receptor blocker treatment group, and piperazine ferulate treatment group. The expression levels of TGF-β1,α-SMA and ColⅠ in the rats were determined by immunohistochemistry assay at 14 days of the experiment. Results Immunohistochemistry results showed that there was slight expression of TGF-β1, α-SMA and ColⅠ in sham-operation group, but the expression in UUO group was significantly increased (P〈 0. 05). The expression of TGF-β1, α-SMA and ColⅠ in angiotensin Ⅱ receptor blocker treatment and piperazine ferulate treatment groups was lower than that in UUO group but higher than that in sham-operation group (P〈0. 05). No significant difference in the expression levels was found between the two treatment groups(P〉0. 05). Conclusion Piperazine ferulate can decrease the expression of TGF-β1,α-SMA and ColⅠ, suggesting it can delay the aggravation of renal tubulointerstitial fibrosis. |
