胆管癌患者血清肿瘤标志物的筛查和鉴定

目的 通过双向凝胶电泳-质谱技术寻找胆管癌患者血清肿瘤标志物.方法 收集8例胆管癌和8例胆管结石患者血清,应用Albumin and IsG Removal kit去除其中的高丰度蛋白等成分,双向凝胶电泳技术分离血清蛋白样本,蛋白分离凝胶采用考马斯亮蓝染色,GPS Explorer~(TM)software凝胶图像分析软件对图像进行数据分析,通过基质辅助激光解析电离飞行时间串联质谱技术进行鉴定.结果 胆管癌和胆管结石患者血清双向凝胶电泳图谱中发现25个差异蛋白质点,20个差异蛋白质点获得成功鉴定,去除相同重复蛋白质后,有8种差异蛋白在胆管结石患者血清中高表达和7种差异蛋白在胆管癌患者血清中高表达.结论 胆管癌与胆管结石患者的血清蛋白中存在较多差异表达的蛋白质,利用双向凝胶电泳技术有望筛选出与胆管癌相关的肿瘤标志物.

Screening and identification of biomarkers in the serum of the patients with cholangiocarcinoma

Objective To search for serum tumor markers from the patients with cholangiocarcinoma and those with cholangiolithiasis using two-dimensional gel electrophoresis (2-DE)-mass spectrometry technique. Methods We chose 8 patients with cholangiocarcinoma patients and 8 with cholangiolithiasis complicated with obstructive jaundice. Albumin and IgG Removal kit was applied to remove albumin components. The serums proteins were isolated using 2-DE technique, and stained using coomassie brilliant blue. The differential expression of proteins was analyzed by using GPS Explorer~(TM) software analysis software. The differentially expressed proteins were screened out and identified through matrix-assisted laser desorption/ionization-time of flight mass spectrometry. Results Twenty-five protein spots were found as differential proteins,in which 20 protein spots were successfully identified. Except for some same proteins, 8 differentially expressed proteins were down-regulated,and 7 differentially expressed proteins were up-reg-ulated in the serum of patients with cholangiocarcinoma. Conclusion There are differentially expressed proteins in the serum from patients with cholangiocarcinoma and those with cholangiocarcinoma. It is hopeful to find the biomarkers related with cholangiocarcinoma using 2-DE technique.
