干扰素-ε诱导K562细胞凋亡的研究

摘    要:
目的:研究应用不同浓度的干扰素(IFN-ε)对K562细胞中野生型p53mRNA和Survivin mRNA表达影响及诱导凋亡作用。方法:应用RT—PCR法检测野生型p53 mRNA和Survivin mRNA的表达。结果:不同浓度的IFN-ε均可抑制Survivin基因的表达,并存在剂量与时间依赖性;可促进野生型p53mRNA的表达上调,但无剂量与时间依赖性。结论:IFN-ε可诱导K562细胞发生凋亡,促进野生型p53 mRNA表达上调和抑制Survivin mRNA的表达可能是其机制之一。

Abstract:
Objective: To study the expression and the induced apoptotic effect of different concentrations of interferon (IFN-ε) to wild-type p53 mRNA and Survivin mRNA of K562 cell. Methods: The expression of wild-type p53 mRNA and Survivin mRNA was detected by RT-PCR. Results: Different concentrations of IFN-ε could inhibit the expression of gene Survivin and occurred in a time-and-dose dependent manner. It could promote wild-type p53 mRNA expression increasing but no time-and-dose dependent manner occurred. Conclusion: It may be one of the mechanisms that IFN-ε may induce K562 cell apoptosis, promote the expression of wild-type p53 mRNA increasing and inhibit the expression of Survivin mRNA.
