筋脉通对实验性糖尿病大鼠坐骨神经AGEs及其受体表达的影响

目的:观察中药复方筋脉通对实验性糖尿病大鼠坐骨神经晚期糖基化终产物AGEs及其受体RAGE表达的影响。方法:采用链脲佐菌素造模后,将大鼠随机分为模型对照组、筋脉通小剂量、中剂量和大剂量组,并设立正常对照组。成模后即开始灌胃给药,筋脉通小、中、大剂量组分别按成人剂量的5、10、20倍给药。模型组和正常组予灌服蒸馏水。检测各组治疗前及治疗后4、8、12、16周的体质量和血糖变化,并测定大鼠机械痛阈,采用免疫组化法测定坐骨神经AGEs及其受体的表达。结果:治疗后正常组大鼠的体质量、血糖与其余各组比较有明显的差异(P<0.01);而模型组及各用药组之间均无明显的差异。机械痛阈结果显示,与正常组比较模型组及各治疗组的机械痛阈值显著降低(P<0.01)。正常组大鼠坐骨神经中AGEs及RAGE表达呈弱阳性,模型组坐骨神经AGEs及RAGE的阳性表达比正常组明显增强,筋脉通各治疗组介于两者之间。IOD值测定显示,与正常组比较,模型组的AGEs及RAGE的IOD值显著升高(P<0.01),各治疗组均较正常组升高,较模型组降低,其中筋脉通中剂量组治疗组明显降低,有显著性差异(P<0.01)。结论:筋脉通能够减少糖尿病大鼠周围神经中AGEs的生成,并下调其受体的异常表达。
