摘    要:
目的 对杜仲抗骨质疏松的药效成分进行初步筛选。方法 采用小鼠成骨细胞MC3T3-E1体外培养模型, 通过MTT法测定细胞增殖, ELISA方法测定碱性磷酸酶(ALP)活性, 观察杜仲中槲皮素、京尼平苷及桃叶珊瑚苷对成骨细胞增殖和分化的影响。结果 槲皮素促MC3T3-E1细胞增殖的有效浓度为10-6 mmol/L, 桃叶珊瑚苷则为10-5 mmol/L;10-4 μmol/L京尼平苷在干预后4 d, 才逐渐显现出促进MC3T3-E1增殖作用。槲皮素(10-5、10-3 mmol/L)、京尼平苷(10-3 mmol/L)和桃叶珊瑚苷(10-3 mmol/L)可增加MC3T3-E1 ALP活性。结论 槲皮素、京尼平苷和桃叶珊瑚苷可促进成骨细胞的增殖和分化, 且作用强度具有浓度相关性和时间相关性, 可能为杜仲抗骨质疏松的药效物质基础。

Abstract:
Objective To screen the pharmaceutical ingredients from Eucommia ulmoides with the function on anti-osteoporosis. Methods The proliferation of osteoblast MC3T3-E1 in mice cultured in vitro was assayed using MTT and ALP activity was assayed using ELISA kit, in order to observe the effect of quercetin, geniposide, and aucubin from E. ulmoides on the proliferation and differentiation of osteoblast. Results The effective concentration of quercetin on MC3T3-E1 proliferation was 10-6 mmol/L and aucubin was 10-5 mmol/L. While geniposide at 10-4 mmol/L could gradually increase the proliferation of MC3T3-E1. The ALP activity of MC3T3-E1 could be increased by quercetin (10-5 and 10-3 mmol/L), geniposide (10-3 mmol/L), and aucubin (10-3 mmol/L). Conclusion Quercetin, geniposide, and aucubin could promote the proliferation and differentiation of osteoblasts, which are the basic materials from E. ulmoides with the function of anti-osteoporosis and is in a concentration-and time-dependent manner.
