雷帕霉素对大鼠坐骨神经再生作用的实验研究

摘    要:
目的探讨雷帕霉素(rapamycin,RAPA)的不同给药时间对大鼠坐骨神经损伤后神经再生和功能恢复的影响。方法健康Wistar大鼠共24只,随机分为4组,制备大鼠右侧坐骨神经挤压伤模型。A组(早期对照组)注射生理盐水,B组(早期用药组)损伤后当即注射雷帕霉素[1mg(/kg.d)],连续应用7d,C组(晚期对照组)注射生理盐水,D组(晚期用药组)损伤后24h注射雷帕霉素[1mg(/kg.d)],连续应用7d,每组各6只。于术后6周进行坐骨神经功能检测(SFI),神经电生理检测及组织形态学观察。结果坐骨神经功能检测(SFI),B组明显优于A组、C组、D组(F=6.51,P0.01)。复合肌肉动作电位(CMAP)的潜伏期(LTP),B组明显慢于A组、C组、D组(F=12.56,P0.01);坐骨神经传导速度(F=20.30,P0.01)和复合肌肉动作电位波幅(F=8.07,P0.01)B组明显优于A组、C组、D组。结论①全身应用雷帕霉素具有神经保护作用,可以加速神经的修复和功能恢复。②早期应用雷帕霉素促进神经再生和功能恢复的效果优于晚期。

Abstract:
Objective To observe the effects of different subcutaneous administration time points of Rapamycin on nerve regeneration and functional restoration after sciatic nerve injury.Methods A total of 24 healthy Wistar rats,divided into four groups,with 6 in each,were prepared to the right sciatic nerve crush injury models.Group A(early control group)received Sodium Chloride,Group B(early treatment group)received Rapamycin(1mg/(kg·d))immediately after injury,continuous application of 7 days,Group C(late control group) received Sodium Chloride and Group D(late treatment group) received Rapamycin [1mg/(kg·d)] 24h after injury,continuous application of 7 days.Results In sciatic nerve function tests(SFI),Group B was significantly better than Group A,Group C and Group D(F=6.51,P0.01).In compound muscle action potential(CMAP) latency(LTP),Group B was significantly slower than Group A,Group C and Group D(F=12.56,P0.01).In nerve conduction velocity(F=20.30,P0.01) and compound muscle action potential amplitude(F=8.07,P0.01),Group B was significantly better than Group A,Group C and Group D.Conclusion ①The systemic application of Rapamycin has neuroprotective effects,which can accelerate nerve repairing and functional recovery.②The early use of Rapamycin to promote nerve regeneration and functional recovery is better than its late application.
