| 摘 要： | 目的通过体外实验研究云南石仙桃氯仿萃取物活性部位对人肝癌细胞株HepG2的细胞增殖抑制作用，并进一步研究其作用机理。方法采用噻唑兰（MTT）比色法测定对HepG2细胞生长增殖的影响；采用流式细胞仪技术和蛋白质电泳技术检测对HepG2细胞的细胞周期及相应的细胞周期蛋白表达的影响。结果云南石仙桃氯仿层活性部位通过下调周期蛋白Cyclin B1及其蛋白激酶p34础的表达将Hep G2细胞阻滞在G2／M期。结论云南石仙桃氯仿层活性部位对人肝癌细胞Hep G2的细胞周期具有阻断作用。

| Abstract: | Objective To investigate the anti-proliferative effect of a fraction of the chloroform extract from Pholidota yunnanesis Rolfe on the HepG2 cells and the mechanism of the induced cytotoxicity. Methods Growth inhibition was analyzed using MTT assay. The effects on the cell cycle of HepG2 cells were detected using flow cytometry. The protein expression was determined using Western blot. Results The fraction showed a considerable cytotoxic activity against HepG2 cells and induced the accumulation of cells in the G2/M phase by down-regulated the protein level of p34^cdc2 and Cyclin B1. Conclusions Our results may contribute to the development of Pholidota yunnanesis Rolfe in cancer treatment. |
