摘    要:目的探讨艾迪注射液中刺五加提取物对斑蝥素的增效、减毒作用。方法采用MTF法考察刺五加提取物对斑蝥素引起的肿瘤细胞(SMMC-7721肝癌细胞)和正常大鼠肾NRK细胞的细胞毒作用;采用流式细胞术检测SMMC-7721和NRK细胞凋亡情况。用斑蝥素制作大鼠急性肾损伤模型,考察刺五加提取物对肾损伤大鼠的保护作用。结果刺五加提取物可增强斑蝥素对SMMC-7721肝癌细胞增殖的抑制作用,降低斑蝥素对NRK细胞的细胞毒作用(P〈0.05或〈0.01);刺五加提取物能增强斑蝥素引起SMMC-7721细胞凋亡,抑制NRK细胞凋亡(P〈0.05或〈0.01)。刺五加提取物可降低斑蝥素所致肾损伤大鼠的尿素氮、肌酐含量,减少尿液中尿蛋白含量(P〈0.05或〈0.01)。结论艾迪注射液中刺五加提取物对斑蝥素有增效、减毒作用。

Abstract:Objective To research the potentiate antitumor and suppress acute nephrotoxicity effects of acanthopanax extract on cantharidin in Aidi injection. Methods MTF essay was used to observe the effect of acanthopanax extract on cytotoxicity caused by cantharidin on cells (SMMC-7721 human hepato- cellular cells and NRK rat kidney cells). Flow cytometry was used to analyze the apoptosis rate of SMMC- 7721 and NRK cells. The protective effects of acanthopanax extract on renal injury caused by cantharidin in rats were projected. Results Acanthopanax extract could not only enhance inhibitional proliferation induced by cantharidin on SMMC-7721 cells, but also reduce cytotoxicity caused by cantharidin on NRK cells. Meanwhile the results of flow cytornetry showed that acanthopanax extract could improve the apopto- sis rate induced by cantharidin on SMMC-7721 cells, as well as reduce the apoptosis rate on NRK cells. Acanthopana~ extract could decrease the contents of blood urea nitrogen, serum creatinine and proteinuria induced by cantharidin in rats. Conclusion Acanthopanax extract has markly effect enhancing and toxic- ity reducing effect on cantharidin in Aidi injection.
