白细胞介素－1β诱导牛脑微血管平滑肌细胞增殖及药物的拮抗作用

研究白细胞介素－１β（ＩＬ－１β）对牛脑微血管平滑肌细胞（ＢＣＳＭＣ）增殖的影响。浓度为５～１００ｎｇ·Ｌ－１的ＩＬ－１β与ＢＣＳＭＣ共同孵育２４ｈ，即可明显诱导ＢＣＳＭＣ的增殖，ＩＬ－１β浓度为５０ｎｇ·Ｌ－１时，增殖率为１７％。欧芹素乙，异欧芹素乙，６－（α，α－二苯基乙酰哌嗪基苯基－４，５－二氢－５－甲基－３（２Ｈ）－哒嗪酮（简称ＤＭＤＰ），６－（α－本基乙酰哌嗪基苯基）－４，５－二氢－５－甲基－３（２Ｈ）－哒嗪酮（简称ＰＭＤＰ）分别在不同浓度可显著抑制ＩＬ－１β所诱导的ＢＣＳＭＣ增殖。

Interleukin-1 beta induced proliferation of bovine cerebromicrovascular smooth muscle cells and antagonism by drugs

The effects of Interleukin-1beta(IL-1β) on the proliferation of bovine cerebromicrovscular smooth cells (BCS MC) were investigated. After 24 h incuba tion with BCSMC IL-1β induced prolifera tion of cultured BCSMC at concentration from 5 to 100 ng· L-1. At 50 ng·L-1,after 24 h incubation with BCSMC, stimulative effect of IL-1β reached 17%. It is found that imperatorin(Imp), isoimperatorin(Isi).6-(αα-diphenylacetylpipezinyl)phenyl-5-methy-4,5-dihydro-3(2H)-pyridazinone (DMDP)and 6-(α-phylly lacetylpipezinyl)phenyl-5-methy-4,5-dihydro-3(2H)-pyridazinone (PMDP0respectively antagonized IL-1β induced prolifera tion of BCSMC at different concentrations.At 10-6 mol·L-1, the inhibitory rated ofImp, DMDP and PMDP were respectively16. 5%,22.44% and 24.09%;At 10-5 mol·L-1, the inhibitory rates of Imp, Isi andDMDP were respectively 38.28%,17,49%and 13.20%.At 10-4 mol· L-1, the inhibitory rates of Imp,Isi and Ver were respectively 30.69%,20. 79% and 31.68%.
