去卵巢骨质疏松大鼠骨组织OPG/RANKLmRNA动态表达研究

摘    要:目的:建立去卵巢大鼠骨质疏松症动物模型,检测造模成功后不同时期大鼠胫骨OPG/RANKLmRNA表达量及血清雌二醇水平,分析OPG/RANKLmRNA表达变化规律。方法:选取3月龄雌性SD大鼠60只随机分为2组即去卵巢组与对照组,每组30只。去卵巢组去除双侧卵巢,对照组仅去除腹腔少许脂肪。分别在造模12周后0个月、6个月、12个月每组随机处死10只,检测不同时期大鼠胫骨OPG/RANKLmRNA表达量及血清雌二醇水平。采用SPSS 17.0软件进行统计,分析数据,得出结论。结果:与对照组相比,去卵巢组胫骨OPGmRNA表达量、血清雌二醇浓度在各时间点上均显著降低(P<0.05),RANKLmRNA表达量则显著增高(P<0.05);去卵巢组0个月与6个月、6个月与12个月比较,胫骨OPGmRNA表达量及血清雌二醇浓度下降,差异具有统计学意义(P<0.05),RANKLmRNA表达量增高,差异具有统计学意义(P<0.05)。结论:造模成功后12个月内,去卵巢大鼠胫骨骨组织OPGmRNA表达量持续减少,血清雌二醇浓度均持续降低,RANKLmRNA持续高表达,OPG/RANKL比值缩小。

Abstract:Objective:To detect OPG / RANKL mRNA expression and serum estradiol concentration at different periods in osteoporotic rats caused by ovariectomy, and to analysis the variation of OPG/RANKL mRNA expression. Methods:Sixty Sprague-Daw female rats aged 3 months were randomized into 2 groups, ovariectomized (OVX) group and sham-operation (SHAM) group, 30 in each group. Rats in OVX group underwent bilateral ovariectomy, while rats in SHAM group only had a small amount of abdominal fat removed. Twelve weeks after the operation was performed, 10 rats of each group were sacrificed at 0,6, and 12 months, respectively, for the measurement of OPG/RANKL mRNA expression and serum estradi ol concentration. Results:OPG mRNA expression and serum estradiol concentration in the OVX group were significantly lower than those in the SHAM group (P〈0.05), while RANKL mRNA expression significantly higher (P〈0.05). In the OVX group, OPG mRNA expression and serum estradiol concentration decreased significantly, from 0 month to 12 month (P〈0.05), while RANKL mRNA expression increased significantly (P〈0.05). Concsion : OPG mRNA expression and serum estradiol concentration reduced continually, and RANKL mRNA expression increased continually in the ovariectomized rats with osteoporosis. The ratio of OPG and RANKL was reduced.
