摘    要:目的:探讨PDCD5在正常前列腺、良性前列腺增生(BPH)及前列腺癌(PCa)组织中的表达特点及其临床意义。方法:通过EnVision法对12例正常前列腺、22例前列腺增生及22例不同病理分级的PCa组织切片进行PDCD5免疫组织化学染色,采用人工阅片评分及计算机辅助染色强度判定2种方法对PDCD5在相应组织中的表达进行定性及定量分析。结果:PDCD5定性及定量检测结果分析显示:正常前列腺与BPH组织中PDCD5表达无显著性差异(P〉0.05),中低危组PCa(Glesaon评分≤7)及高危组PCa(Glesaon评分〉7)组织中PD—CD5表达与正常前列腺与BPH均存在显著性差异(P〈0.01),并且两组间PDCD5表达亦存在显著性差异(P〈0.05)。结论:PDCD5在正常前列腺及BPH组织中近似地呈现较强的阳性表达。PDCD5在PCa组织中的表达较正常组织及BPH均有显著下降,并且随着PCa恶性程度上升,PDCD5的表达进一步下调,提示PDCD5可能在不同程度上参与了PCa发生、发展过程中腺细胞凋亡的调控。

Abstract:Objective:To assess the expression of PDCD5 in tissues of normal human prostate(NP), BPH and prostate cancer(PCa). Methods:The expression of PDCD5 in 12 NP,22 BPH and 22 PCa samples were tested by EnVision immunohistochemical methods. Positive expression rates and intensity of PDCD5 protein were investigated by observing under microscope and analyzing with computer imaging technique. Results: The statistical outcomes on the basis of quantitative analysis and qualitative analysis showed, there was no significant difference be tween the expression of PDCD5 in NP and BPH tissues(P〉0. 05). Low/ middle-risk PCa and high-risk PCa group comparing with NP and BPH group indicated obvious statistical significance(P〈0.01), what's more, the expression of PDCD5 in Low/middle-risk and high-risk PCa has obvious differences(P〈0.05). Cunclusions:PDCD5 expressed strongly both in NP and BPH tissues with no difference. Adversely, PDCD5 expression was obvious lower in PCa than in NP and BPH and was downregulated further with the increase of Gleason sum, which indi cates PDCD5 may be an important apoptosis regulating factor in the occurrence and development of PCa.
