摘    要:
目的:研究阴离子交换树脂对Streptomyces roseosporus发酵液中达托霉素的纯化工艺。方法:通过比较不同种类的阴离子交换树脂,以树脂对达托霉素的吸附量、洗脱率、纯度为考察指标综合评价。结果:筛选出对发酵液中达托霉素吸附较高,并具有较高回收率,杂质吸附较低的阴离子交换树脂330。确定最佳吸附条件为:吸附方式为静态吸附,吸附温度为15℃~30℃,吸附液的pH8.0~8.5,饱和吸附时间为10 h;最适洗脱条件为:洗脱方式为静态洗脱,洗脱剂为0.8 mol.L-1 NaCl水溶液,洗脱体积为12倍树脂体积,洗脱时间为8 h。结论:确定了阴离子交换树脂330对达托霉素的纯化工艺,在优化条件下,发酵液中达托霉素的纯度由2%提高到了60%,回收率达87.5%。

Objective: To find the best purification process of daptomycin from Streptomyces roseosporus fermentation broth.Methods: The absorption behavior of daptomycin in fermentation broth on several anion-exchange resins was studied and the absorption,desorption ratio and purity were used as markers to screen the best method of extraction and adsorption conditions of anion-exchange resins.Results: Resin 330 was chosen to separate daptomycin due to its high recovery rate and low absorption for impurities.The optima...
