| 摘 要： | 目的 观察葛根素对T诱导的血管平滑肌细胞(VSMC)增殖的影响。方法 建立凝血酶(T)诱导的大鼠血管平滑肌细胞增殖模型,以细胞计数法和流式细胞仪DNA含量测定的方法观察T及葛根素对VSMC增殖和DNA合成的影响。结果 T对VSMC有明显促增殖作用,促增殖效应在2 4小时末达峰,且T浓度在0 . 1U/L～1 . 0U/L之间有剂量依赖关系;葛根素呈剂量依赖性地抑制T诱导的细胞增殖与DNA合成。结论 葛根素能抑制T诱导的VSMC增殖。 |

| Abstract: | Objective To observe the effect of puerarin on proliferation of vascular smooth muscle cells (VSMC) induced by thrombin(T) in rats. Methods Cell number and cell cycle analysis using flow cytometry were adopted as two different indicators of proliferation of VSMC. Results T can significantly increase cell number of VSMC with the peak of proliferation at 24h. The effect of T is in a dose-dependent manner (0.1U/L-1.0U/L). Puerarin 1.5×10 -5 mol/L-1.5×10 -3 mol/L significantly suppresses the stimulation proliferation of VSMC and DNA synthesis induced by T. Conclusions Puerarin can suppress the proliferation and DNA synthesis of VSMC induced by T. |
