| 摘 要： | 目的观察角质细胞生长因子(KGF)对人卵巢上皮性癌细胞系CAOV3增殖及酪氨酸蛋白激酶(TPK)活性及表达的影响。方法培养人卵巢上皮性癌细胞系CAOV3,用四甲基偶氮蓝(MTT)比色法测定不同浓度KGF对细胞增殖的影响;通过免疫沉淀法纯化蛋白并用TPK试剂盒测定TPK活性;免疫印迹法(Western-blot)测定TPK表达。结果实验组CAOV3细胞增殖率、TPK活性及表达均大于对照组(P<0·05)。结论KGF可以促使人卵巢上皮性癌细胞系CAOV3增殖,机制与其增强TPK活性及表达有关。

| Abstract: | Objective To observe the effects of keratinocyte growth factor (KGF) on the proliferation of CAOV_3 cell line and the activity and expression of tyrosine protein kinase (TPK).Methods CAOV_3 cell line of ovarian cancer was cultured,cell line proliferation was measured by flow cytometry and MTT assay;the activity of TPK was measured by immune-precipitation meathod;and the expression of TPK was assessed by Western blot.Results The proliferation of CAOV_3 cell line,activity and expression of TPK were higher in the experiment group than those in the control group(P<0.05).Conclusion KGF can stimulate the proliferation of CAOV_3 cell line,and the mechanism is related with the activity and expression of TPK. |
