温度对低密度脂蛋白胆固醇测定的影响

摘    要:目的:比较不同温度以聚乙烯硫酸(PVS)沉淀法分离血清脂蛋白的效果,并观察温度对低密度脂蛋白胆固醇(LDL-C)测定值的影响。方法:选取6℃,20℃,30℃条件下,用PVS作沉淀剂对37份血清标本及低密度脂蛋白胆固醇定值血清进行沉淀分离,取上清液测定胆固醇含量,用定值血清原液做标准液计算LDL-C值。结果:用定值血清原液做标准计算LDL-C值,37份标本均值6℃比20℃高5.56%,20℃比30℃高6.83%;用相同温度沉淀处理的定值血清上清液做标准计算LDL-C值,37份标本均值6℃比20℃高3.11%,20℃比30℃高1.69%。结论:温度对PVS沉淀分离血清脂蛋白的效果有影响,使用同条件沉淀处理的定值血清做标准测定LDL-C值较好。

Abstract:Objective:To compare the separating efficiency of serum lipoprotein by polyvingel sulfate(PVS) sedimentation method under different temperatures and observe effect of the temperatures on the measurements of low density lipoprotein cholesterol(LDLC). Method:Utilizing polyvinyel sulfate as sedimentary agent,37 serum samples and serum with known concentration of LDLC were precipitated and cenfrifuged under 6,20 and 30 separatedly,supertanants were taken for LDLC determination.The values determined from serum with known LDLC concentration under different temperatures were used as standard values for sample LDLC Calculations.Results:The mean LDLC values of the 37 serum samples calculated by using LDLC values defermined from serum with known LDLC as standands were 5.56% ligher under 6 than under 20,and 6.83% ligher under 20 than under 30.The mean LDLC value of 37 serum, calculated by using value of known LDLC determined under one temperature,was 3.11% higher under 6 than that under 20,which was again higher (1.69%) than that under 30.Conclusion:The results indicate that serum lipoprotein cholesterol determination by using PVS sedimentation method is affected by temperature.It is recomanded that it is better to use the same precipitation condition for both standard LDLC determination and sample LDLC determination.
