蓝莓花青素与白藜芦醇复配对老龄小鼠抗衰老作用的评价

摘    要:目的:探讨蓝莓花青素与白藜芦醇复配对老龄小鼠的抗衰老作用。方法:采用自然衰老动物模型,将蓝莓花青素和白藜芦醇复配连续灌胃老龄小鼠30 d,以幼龄小鼠为空白对照组,通过测定各组小鼠血清中的超氧化物歧化酶(superoxide dismutase,SOD)活性、丙二醛(malondialdehyde,MDA)含量、总抗氧化能力(total antioxidant capacity,T-AOC)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力以及小鼠的主要脏器指数,综合评价蓝莓花青素和白藜芦醇复配后高、低剂量组(200 mg·kg-1+20 mg·kg-1、100 mg·kg-1+10 mg·kg-1)对老龄小鼠体内抗衰老作用。结果:与老龄对照组比较,蓝莓花青素与白藜芦醇复配高、低剂量组的小鼠血清SOD活性显著提高(P<0.05),并能显著降低血清中MDA的含量(P<0.05),表明复配样品具有良好减缓老龄小鼠衰老进程的活性;进一步测定小鼠血清中的抗氧化指标,结果显示蓝莓花青素与白藜芦醇复配组小鼠血清T-AOC和GSH-Px活性均有显著提高(P<0.05)。除此之外,与老龄对照组比较,蓝莓花青素与白藜芦醇复配组可显著提高老龄小鼠的胸腺指数(P<0.05)。与白藜芦醇组比较,复配的高剂量组小鼠血清总抗氧化能力有显著提高(P<0.05);在其他方面没有表现出明显的协同增效作用。结论:蓝莓花青素与白藜芦醇复配可能通过提高老龄小鼠胸腺免疫器官的功能,并改善其体内抗氧化酶系统的活性,达到延缓衰老的作用。其具体协同增强作用机制有待进一步研究。

Abstract:OBJECTIVE To evaluate anti-aging effect of Gardenblue blueberry anthocyanin and resveratrol in senile animals. METHODS Naturally senile Kunming mice were gavaged with Gardenblue blueberry anthocyanin and resveratrol once a day for 30 days at our prescribed dose. Gardenblue blueberry anthocyanin and resveratrol at high (200 mg·kg-1+20 mg·kg-1) and low (100 mg·kg-1+10 mg·kg-1) doses were evaluated comprehensively in vivo, by determining the superoxide dismutase activity (SOD), the malondialdehyde content (MDA), the total antioxidant capacity (T-AOC) and glutathione peroxidase activity (GSH-Px) in mice serum, as well as main organ index of mice compared with the blank control group of young Kunming mice. RESULTS Compared with control group of naturally senile mice, the experiment groups of Gardenblue blueberry anthocyanin and resveratrol at high and low doses increased SOD activity in mice serum (P<0.05), as well as reduced content of MDA in serum (P<0.05). The result indicated that compound samples could reverse aging process. Further, compared with control group of naturally senile mice, T-AOC and GSH-Px of the high and low dose groups were increased (P<0.05). In addition, thymus index of compound samples was higher than that of the model group. Compared with resveratrol group, the total antioxidant capacity of mice in the high dose group was significantly increased (P<0.05), and there was no obviously synergistic effect in other aspects. CONCLUSION Gardenblue blueberry anthocyanin and resveratrol can improve the activity of antioxidant enzymes in vivo through enhancing the function of immune organs to delay aging. The mechanism needs further study on the mechanism of the specific synergy.
