胰腺癌组织中差异表达蛋白的筛选与鉴定

摘    要:目的:筛选并鉴定胰腺癌组织中的差异表达蛋白,并探讨其临床意义。方法:应用双向凝胶电泳技术对12对胰腺癌组织和癌旁组织样品进行分析,筛选出差异表达蛋白质;应用MALDI-TOF-MS/MS技术对差异表达蛋白质进行鉴定。结果:优化的胰腺组织双向凝胶电泳技术具有良好的分辨率和可重复性;应用双向凝胶电泳技术筛选出胰腺癌组织中30个差异表达蛋白质;应用MALDI-TOF-MS/MS技术鉴定了胰石蛋白、钙囊素、热休克蛋白B6等24个蛋白质,其中15个蛋白质在胰腺癌组织中表达上调,9个蛋白质表达下调。结论:双向凝胶电泳、生物质谱等蛋白质组学技术在胰腺癌的基础研究中有重要价值和应用前景,有助于筛选蛋白标志物和阐明分子发病机制。胰腺癌组织中有多个异常表达蛋白质,这些蛋白质与胰腺癌的发生相关,可能成为胰腺癌蛋白标志物和药物治疗的靶蛋白。

Abstract:Objective To screen and identify differential expression proteins from pancreatic carcinoma and neighboring normal pancreatic tissues and to study their clinical significance. Methods Two-dimensional gel electrophoresis(2-DE) was applied to separate differential expression proteins among 12 pairs of cancer and their corresponding neigboring pancreas. MALDI-TOF-MS/MS was then applied to identify these differential expression proteins. Results The improved 2-DE map demonstrated effectiveness good reproducibility. Twelve pairs of cancer and corresponding neigboring pancreas were analyzed by 2-DE,and 30 proteins were found to be differently expressed between cancer and normal pancreatic tissues. Furthermore,24 proteins were identified by MALDI-TOF-MS/MS,including pancreatic stone protein,calgizzarin,and heat shock protein B6;15 proteins were up-regulated in cancer tissues,while 9 proteins were down-regulated. Conclusions Proteomic technologies,such as 2-DE and mass spectrometry,had clinical significance and important applications in the study of pancreatic cancer. These technologies could help to screen biomarkers and to elucidate mechanism. Many proteins were abnormally expressed in the tissues of pancreatic cancer. These proteins had relationships with pancreatic cancer and could be biomarkers or target proteins in the future.
