摘    要:目的研究实验性肝纤维化过程中明胶酶A(mmp-2)酶原蛋白表达的动态变化.方法建立四氯化碳大鼠肝纤维化模型,采用Western印迹法和免疫组化方法,对肝纤维化不同阶段肝内mmp-2的酶原蛋白的表达水平与分布进行了检测.结果在正常大鼠肝组织中有mmp-2的酶原表达.在肝纤维化发生发展过程中mmp-2酶原蛋白表达水平逐渐增强,至肝纤维化中期到达最高峰,到肝纤维化晚期及肝硬变阶段其表达较中期下降,但仍高于正常对照水平.正常肝组织内,mmp-2的表达主要位于汇管区的胆管壁和毛细血管壁及肝窦壁的间质细胞;纤维化时,增生的纤维间隔附近亦有较高表达.结论 mmp-2在肝纤维化的发生、发展直至肝硬变形成过程中发挥着重要作用.

Abstract:AIM To investigate the dynamic alteration of gelatinase A (metalloproteinase-2, mmp-2) proenzyme protein expression in the process of experimental liver fibrosis. METHODS The carbon tetrachloride-induced hepatic fibrosis rat models was used. mmp-2 proenzyme protein expression in the different stages of liver fibrosis was measured by quantitative analysis of Western blot and immunohistochemical technique.RESULTS mmp-2 proenzyme protein expression could be detected in normal rat liver. In the pathogenesis and development of liver fibrosis, mmp-2 proenzyme protein expression level was increased gradually, the expression rate being highest at an intermediate stage of fibrosis. And it decreased gradually during the late liver fibrosis and at stage of liver cirrhosis, but remained relatively high levels above normal. The expression of mmp-2 was located in sinusoidal cells, biliary ducts and capillary endothelial cells in the space of Disse, it was more intense around fibers in fibrous septa in liver cirrhosis. CONCLUSION mmp-2 might participate in pathogenesis of liver fibrosis and cirrhosis as an important role.
