黄芪对高糖高脂环境下内皮细胞与系膜细胞相互作用的影响

摘    要:
目的探讨黄芪对高糖高脂环境下内皮细胞与系膜细胞间相互作用的影响。方法内皮细胞与系膜细胞共培养和系膜细胞单独培养,分为对照组、甘露醇组、高糖高脂组、黄芪组,培养24h后,ELISA法检测各组细胞上清液IV型胶原(colIV)、纤维连接蛋白(Fn)的含量。结果在高糖高脂环境下,共培养和单培养细胞上清液ColIV、Fn含量升高(均P〈0.05),共培养上清液C01IV、Fn较单培养升高(均P〈0.05);黄芪组colIV、Fn含量较高糖高脂组下降(均P〈0.05)。结论高糖高脂环境下,系膜细胞和内皮细胞存在异常的相互作用,这种作用能促进ColIV、Fn产生。黄芪通过抑制高糖高脂条件下内皮细胞与系膜细胞间的相互作用,减少ColIV、Fn的含量,发挥肾脏保护作用。

Effect of Astragalus Membranaceus on the Interaction of Mesangial Cells and Endothelial Cells

Abstract:
Objective To study the effect of astragalus membranaceus on the interaction of mesangial cells and endothelial cells in the media of high glucose and lysophosphatidylcholine. Methods The model of intercellular interaction between endothelial cells and mesangial cells of diabetic nephropathy was established and divided into 4 groups: control, mannitol, high glucose and lysophosphatidylcholine,intervented with astragalus membranaceus. Endothelial cells and mesangial cells were co-cultured in DMEN with or without astragalus membranaceus in high glucose and lysophosphatidylcholine media up to 24 hours. The level of collagen IV and fibrocentin were determined by enzyme linked immunosorbent assay in the culture media. Results The level of collagen IV and fibrocentin of high glucose and lysophosphatidylcholine group was markedly higher than that of control group in co-culture and the ordinary monolayer cell culture (P 〈0.05). Intervented by high glucose and lysophosphatidylcholine, the level of collagen IV and fibrocentin of co-culture was much higher than that of the ordinary monolayer cell culture (P 〈 0.05). The levels of collagen IV and fibrocentin of the group intervented by astragalus membranaceus was lower than that of high glucose and lysophosphatidylcholine (P〈0.05). Conclusions Exposed to high glucose and lysophosphatidylcholine, endothelial cells and mesanglal cells can interact with each other to promote collagen IV and fibrocentin. Astragalus membranaceus can inhibit the interaction of endothelial cells and mesangial cells in the media of high glucose and lysophosphatidylcholine to decrease the level of collagen 1V and fibrocentin and has protective effect on the kidney.
