达纳康对鼠脑缺血-再灌注后ICAM-1的影响

摘    要:
目的:观察达纳康对脑血管内皮的保护性作用及脑血管扩张作用。方法:选用非开颅线栓法造成鼠大脑中动脉缺血,缺血2h后行再灌注术。分别检测再灌注不同时点下的细胞黏附分子-1(ICAM-1)、NO。结果;在不同再灌注时限,对照级3个亚组间ICAM-1有显著变化,尤其是再灌注12h组明显高于再灌注24h和48h组。在相同再灌注时间下,对照组ICAM-1高于服药组,NO低于服药组。结论:达纳康对ICAM-1有明显的抑制作用。并且通过NO释放的增加,增加血管张力,减轻再灌注的又一次损伤。

Abstract:
Objective: To investigate the beneficial effects of Danakan on cerebral endothelia cells and distension of cerebral ves-sels. Methods: Animal models with middle cerebral artery occlusion (MCAO) were made with thread without craniectomy. After 2h of MCAO, the thread was pulled out and reperfusion was performed. Some of them were treated with Danakan as therapeutic group and others were not treated as control group. The nitric oxide (NO) and intercellular adhesion molecule-1 (ICAM-1) contents at different reperfusion lime were determined. Results; In control group, the ICAM content at 12h was significantly higher than those at 24h and 48h. Al the same reperfusion time, the ICAM-1 content in control group was higher than that in therapeutic group, but the NO content in control group was lower than that in therapeutic group. Conclusion: Danakan markedly inhibiles the expression of ICAM-1 and stimulates the expression of NO, which may protect he-mangiectasia and reduce ischemia-reperfusion damage.
