内皮抑素和平阳霉素对血管内皮细胞的作用

目的：比较重组人血管内皮抑制素（rHES）、平阳霉素（PYM）对血管内皮细胞（HUVEC）的作用效果,探讨相关作用机制,以揭示rHES应用于增生期血管瘤治疗的可行性。方法：将不同浓度rHES、PYM分别作用于HUVEC细胞,以MTT法比较两药作用24h、48h后的细胞抑制率,苏木精-伊红（HE）染色光镜下观察细胞形态变化,用免疫组化法检测细胞内血管内皮生长因子（VEGF）阳性表达的差异,流式细胞术分析细胞凋亡。结果r：HES、PYM作用后的吸光度值（OD值）均随浓度增大而减小,VEGF免疫组化染色细胞灰度值均增大,流式细胞仪测得两药均会引起细胞凋亡,联合使用表现出协同效应。光镜下可见：rHES作用后细胞形态明显改变,可看到大量凋亡细胞,PYM作用后大量细胞悬浮,贴壁细胞数量明显减少。结论r：HES、PYM均能促进HUVEC细胞凋亡,联合使用有协同作用。

Effects of endostatin and pingyangmycin on vascular endothelial cell

Objective To compare the effects of recombinant human endostatin（rHES）and pingyangmycin（PYM） on vascular endothelial cells,（HUVEC）,and discuss the relevant mechanisms,and reveal the rHES used in the treatment of proliferative hemangioma feasibility.Methods After HUVEC cells were treated with different concentrations of rHES and PYM for 24hrs and 48hrs,the inhibition rates were compared respectively by MTT method,the changes in cell morphology were observed under light microscope by Hematoxylin-eosin（HE） staining,the differences of vascular endothelial cell growth factor（VEGF） in cell were detected by immunohistochemistry,and the apoptosis was analyzed by flow cytometry.Results After were treated by rHES,PYM,the absorbance value（OD value） were decreased with increasing concentration,the cell gray levels were increased by VEGF immunohistochemical staining,apoptpsis was measured by flow cytometry which was induced by both of the two drugs,the combination of the two drugs showed synergy effects.Under the light microscope,the morphological changes of cells that were treated with rHES which was significant,also it could be seen as a large number of apoptotic cells,while the suspended cells increased and the number of adherent cells decreased largely after treating with PYM.Conclusion rHES and PYM both could increase apoptosis in HUVEC cells,also combination using of them had synergistic effects.
