滋阴清热法对抑郁症模型大鼠脑内单胺类神经递质、脑源性神经营养因子和酪氨酸激酶B的影响

摘    要:目的:研究滋阴清热法对抑郁模型大鼠的影响,揭示知母的抗抑郁作用机理。方法:分别从分子水平及病理改变对比3组间单胺类神经递质(5-HT、NE)、脑源性神经营养因子(BDNF)和酪氨酸激酶B(TrkB)在抑郁症大鼠模型脑内表达情况。结果:光镜结果提示滋阴清热治疗可使大脑外层皮质区神经元细胞、锥体细胞数量明显增多,细胞核核仁明显未见病理性变化,锥体细胞层毛细血管未见出血;免疫组化结果提示生理盐水组及滋阴清热组大鼠5-HT、NE、BDNF、TrkB的表达较正常组差(P〈0.05),但生理盐水组和滋阴清热组的5-HT表达上差异无统计学意义(P〉0.05)。结论:滋阴清热中药知母具有一定的神经保护作用,但其具体机制仍需要进一步深入的探索。

Effect of Nourishing Yin and Clearing Heat Therapy on Monoamine Neural Transmitter,Brain-deried Neurotrophic Factor,Tyrosine Kinase B in Depression Rat Model's Brain

Abstract:Objective:To reveal the mechanism of anti - depression by nourishing yin and clearing heat therapy (NYCHT) and to study the effects in depression rat model' changes from molecular level and pathology. Methods :80 SD male rats conforming to similar score of Open - field test were made mode by unpredictable chronic stresses methods and were randomly divided into physiological saline group and NYCHT group ( n = 40 ) , another 40 SD male rats as control group was observed at the same time. After 2 weeks, the expression of Monoamine neural transmitter (5 - HT, NE), Brain -deried neurotrophic factor (BDNF) ,Tyrosine kinase B( TrK B) in depression rat models' brain were assayed and comparatively analyzed. Results : Nerve cell of pallium and pyramidal cell significantly elevated and karyon nucleolus unconspicuous pathological changing as well as normal capillary of pyramidal cell layer was observed in NYCHT group. Analysing of immunohistochemistry : the expressions of 5 - HT, NE, BDNF and TrK B in rats' brain of saline group and NYCHT group were lower than that in control gruop ( P 〈 0.05 ), there was no significant difference on the expression of 5 - HT between saline group and NYCHT group in statistics ( P 〉 0. 05 ). Conclusion : Anemarrhena asphodeloides Bge for NYCHT has neuroprotection to some extent,however,its mechanism should need further explortion. Key words:
