大孔吸附树脂分离纯化枇杷花总黄酮工艺研究

摘    要:
目的:筛选一种适合分离纯化枇杷花总黄酮的大孔吸附树脂并确定其最佳工艺参数。方法:以总黄酮的吸附率与解析率为指标,从D101、AB-8、DM301、S-8四种大孔吸附树脂中筛选出最佳树脂,并通过单因素考察该树脂分离纯化总黄酮的最佳工艺条件。结果:DM301树脂对枇杷花总黄酮具有良好的吸附分离性能,最佳工艺参数为:上样浓度12.5 mg/mL,上样流速2 mL/min,上样pH 5.0,树脂最大上样量60 mg/g;最佳洗脱剂50%乙醇,洗脱速率3 mL/min,洗脱剂用量10 BV。结论:在所确定的工艺条件下,DM301树脂分离纯化枇杷花总黄酮效果良好,以Al(NO3)3-NaNO2-NaOH比色法计,产物经干燥后总黄酮质量分数最高可以达到80%以上。

Studies on Separation and Purification of Total Flavones from Loquat Flower with Macroreticular Resin

Abstract:
Objective:To screen the best macroporous adsorptive resin for separation and purification of Loquat flower flavonoids and to establish the purification process parameters. Methods:Using macroporous adsorption resin adsorption rate and desorption rate as the indexes, the best resin was screened out for enriching Loquat flower flavonoids from D101, AB- 8 ,DM301 and S -8 resin. The optimal process conditions were investigated through single factor test. Results: DM301 resin showed good adsorption and separation property for Loquat flower flavonoids. The detailed process conditions were as follows : sample concentration was 12.5 mg mL-1 ; sample loading rate was 2.0 mL min-1; pH was adjusted to 5.0 ; the largest sample load was 60 mg g-1 Eluent concentration was 50% with elution rate of 3 mL min-1 and the dosage was 10 BV. Conclusion:DM301 type resin showed good purification effect for Loquat Flower flavonoids in the de- fined conditions. The content of total flavonoids in the dried matter could increase to 80% above measured by AI( NO3 )3 - NaNO2 - NaOH colorimetric method.
