响应曲面法优化吉祥草皂苷提取工艺

摘    要:目的:优化吉祥草皂苷提取工艺。方法:采用紫外分光光度法测定皂苷含量,以提取率为考察指标,用响应曲面法优化最佳提取工艺。结果:吉祥草皂苷的最佳提取工艺为:料液比为(g∶mL)20.39∶1,81.24℃条件下,水浴回流提取2次,每次79.76 min。用该工艺条件进行提取,吉祥草皂苷得率为1.36﹪。吉祥草药材皂苷含量在2.743~13.825 mg/g之间,平均为1.657 mg/g。结论:该最佳提取工艺稳定可靠,简单易行,能够用于吉祥草皂苷提取。

Abstract:Objective : To optimize the extracting process of saponin in Reineckia carnea. Method : Investigating the influ- ence of three factors, namely extracting temperature, extracting time and ratio of solid - liquid, on the yield of total sapo- nins in Reincckia carnca with Box - Behnken response surface methodology, saponin content was determined by ultraviolet spectrophotometry. Result : The temperature had a significant influence on the yield of saponin, flowed by extracting time, while the ratio of solid - liquid was less. And the productivity was the highest and reached 1.36% under the optimal pro- cessing parameters of the temperature 82 ~C ,time 80 min and rate 1 : 21. The contents of steroidal saponin were 2. 743 13. 825 mg/g,averagely 9. 088 mg/g in Reincckia carnca. Conclusion:The optimal extracting process is steady, reliable and easily for using, and can be used for extracting saponins in Reineckia carnea.
