川芎嗪对IgA肾病大鼠的肾脏保护作用及TGF—β表达的影响

摘    要:目的:研究川芎嗪对IgA肾病(IgAN)大鼠肾脏的保护作用及对TGF—β表达的影响。方法:建立IgA肾病大鼠模型,随机分成模型组、地塞米松组、川芎嗪组,另设正常对照组;光镜观察肾组织形态,RT-PCR检测肾组织的TGF-βmRNA、表达,ELISA法测血清TGF—β水平。结果:光镜下川芎嗪治疗组IgAN大鼠的肾组织病理损害减轻。RT—PCR结果显示地塞米松与川芎嗪下调TGF-βmRNA,与模型组相比差异有统计学意义(P〈0.01);ELISA结果提法明地塞米松与川芎嗪显著降低血清TGF—β水平,与模型组相比差异均有统计学意义(P〈0.01)。结论:川芎嗪能明显改善IgAN大鼠肾组织的病理损害。川芎嗪能够下调肾组织TGF-βmRNA、TGF—β蛋白水平,这可能是川芎嗪治疗IgA肾病的作用机制之一。

Abstract:Objective :To explore the effect and mechanism of tetramethylpyrazine (TMP) on renal protection and TGF - β expression of rats with IgA nephropathy. Methods:After establishing the rat model of IgA nephropathy,female Sprague- Dawley(SD)rats were divided into lgA nephropathy model group( group B),dexamethasone group (group C)and TMP group( group D). The control group (group A) was set up too. The renal tissue morphology was observed with light microscope ; mRNA expression levels of TGF -β were investigated with RT - PCR, and the serum levels of TGF - β was evaluated with ELISA. Results:Compared with group B,the damage of renal tissue was reduced in group D. The mRNA expression of TGF - β in rat nephridial tissue of group C was significantly lower than that of group B ( P 〈 0. O1 ) . The mRNA expression of Nephrin in rat nephridial tissue of group C was significantly elevated than that of group B (P 〈 0.01 ). The serum levels of TGF -β protein in group C and D were lower than those in B group( P 〈 0.01 ). Conclusion: TMP can reduee the renal tissue damage of rat model of IgA nephropathy. TMP shows the ability of down - regulating the expression of TGF - 13 mRNA and TGF - 13 protein, which may explain the meehanism of TMP in clinical treatment of IgA nephropathy.
