摘    要:目的:建立两对引物-聚合酶链反应(PCR-CTPP)技术改进体系-两步法PCR-CTPP,研究其在MDR1基因SNP的快速分型中的应用价值。方法:采用两步法PCR-CTPP技术对158例的癫痫患者MDR1基因rs3789243和rs2235046进行检测分型,并以DNA测序验证基因分型结果。结果:通过两步法PCR-CTPP得到的基因分型结果与DNA测序得到的基因分型结果完全一致。结论:改进的两步法PCR-CTPP技术是一种可靠、快速的对基因SNP分型检测方法,能在普通实验室广泛应用,有助于基因SNP的分型研究。

Abstract:Objective:To evaluate the value of improved two-step polymerase chain reaction with confronting two-pair primers(PCR-CTPP)for quick genotyping MDR1 gene single nucleotide polymorphism(SNP).Methods:MDR1 gene polymorphism rs3789243 and rs2235046 were detected by two-step PCR-CTPP in 158 epileptics and genotyped with DNA sequence analysis.Results:The results of genotyping by two-step PCR-CTPP were consistent with those conducted by DNA sequence analysis.Conclusion:The improved two-step PCR-CTPP appears a fast and reliable method for gene genotyping and may be recommended in wider general laboratory use in research of gene SNP genotyping.
