摘    要:目的:建立测定人血清中丙戊酸钠浓度的方法。方法:血样采用2-溴-对硝基苯乙酮衍生化,以高效液相色谱法进行测定。色谱柱为Shimpack C18,流动相为甲醇-水(80∶20),流速为0.9 mL·min-1,检测波长为265 nm,柱温为30 ℃,内标为环己烷羧酸。结果:丙戊酸钠血药浓度在10~150 mg·L-1范围内线性关系良好(r=0.999 7,n=6),最低检测浓度为1 mg·L-1;方法回收率为99.45%~103.24%,萃取回收率为98.12%~100.37%,日内与日间RSD均<8%(n=5)。结论:本方法快速、简便、准确,适用于临床丙戊酸钠的血药浓度监测。

Abstract:OBJECTIVE:To establish the method for the concentration determination of sodium valproate in human serum. METHODS: The samples were derivatized with 2-Bromo-4′ -nitroacetophenone, and HPLC method was adopted. The chromatographic column was Shimpack C18(150 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-water (80 ∶ 20) at flow rate of 0.9 mL·min-1. The colum temperature was 30 ℃ and the detection wavelength was set at 265 nm. The cyclohexanecarboxylic acid was used as internal standard. RESULTS:The linear range of sodium valproate was 10~150 mg·L-1(r=0.999 7,n=6). The minimum detectable concentration was 1 mg·L-1. The average recovery of relative and extraction was 99.45%~103.24% and 98.12%~100.37%. The RSD of intra-day and inter-day were less than 8%(n=5). CONCLUSION: The method is rapid, simple and accurate, which can be used for serum concentration monitoring of sodium valproate.
