紫杉醇温敏纳米胶束对人胃癌裸鼠移植瘤的靶向治疗作用

摘    要:目的:探讨紫杉醇温敏纳米胶束靶向治疗人胃癌裸鼠移植瘤的疗效。方法:建立裸鼠BGC-823胃腺癌动物模型48只,随机分为紫杉醇热疗和非热疗组、紫杉醇温敏纳米胶束热疗和非热疗组、温敏纳米胶束热疗和非热疗组、单纯热疗组和等渗盐水组,每组6只,采用尾静脉给药方式,并43℃恒温对热疗组肿瘤部位进行热导向化疗,动态观察并测定肿瘤的生长。结果:紫杉醇温敏纳米胶束热疗组对人胃癌的移植瘤有显著抑制作用,肿瘤体积和瘤重抑瘤率分别为83.9%和81.3%,显著高于紫杉醇温敏纳米胶束非热疗组(43.6%和39.5%)和紫杉醇注射液组(49.6%和54.2%)(P〈0.05),而且对正常组织的毒副反应明显减轻。结论:温敏纳米胶束具有良好的肿瘤抑制效果,作为一种新型的药物载体,显示出良好的应用前景。

Abstract:Objective: To study the curative effect of paclitaxel thermosensitive nano-micelle(PTN) on human gastric cancer xenografts in nude mice.Methods: Human gastric cancer cell line BGC-823 was implanted into 48 nude mice.Drugs were injected through the caudal vein in 8 different groups with established experimental models.Tumor growth was monitored once every other day.Constant temperature was applied to tumor in the targeting groups. Results: The growth speed of tumor in the group of paclitaxel thermosensitive nano-micelle thermotherapy was significantly slower than the other groups.The rates of tumor restrain in tumor weight and tumor volume of the paclitaxel thermosensitive nano-micelle thermotherapy group were 83.9% and 81.3%,respectively,remarkably higher than those of the paclitaxel injection group(49.6% and 54.2%) and the paclitaxel thermosensitive nano-micelle group(43.6% and 39.5%)(P< 0.05).The side-effect and toxicity in the paclitaxel thermosensitive nano-micelle thermotherapy group were significantly less than those in the other groups.Conclusion: Paclitaxel thermosensitive nano-micelle can inhibit the growth of tumor,which indicates a favorable foreground for its clinical application.
