诱导成骨修复大鼠颅骨缺损

摘    要:本文定性、定量地考察了骨基质明胶、脱钙骨和深冻骨促进SD大鼠颅骨缺损修复的作用。结果表明它们均有不同程度的诱导成骨作用,其中骨基质明胶的作用最佳,脱钙骨次之。术后12周,骨基质明胶组的骨缺损已完全愈合,脱钙骨组接近愈合。本文还讨论了诱导成骨的机制,认为骨形态形成蛋白(BMP)是促进骨缺损愈合的主要生物活性物质:巨噬细胞可能与整个诱导过程的启动有关。

Experimental study on the enhancement of rat skull defects' repair by induced osteogenesis

Abstract:The repairing capacity of bone matrix gelatin (BMG), decalcified bone matrix ( DBM ) and deep frozen bone(DFB ) were tested in experimental defects on skulls of rats, in which trephine defects of 7mm in diameter were made. The healing of the defects were judged by roentgenographic, histological and atomic absorption spectrophotometric methods. Healing of defects was most effective with BMG(P< 0.01), DBM came the next, while the capability of bone induction was very weak in DFB. In BMG and DBM groups, skull defects were completely healed in 12 weeks after the operation, but in DFB group healing was incomplete. The control group failed to heal spontaneously.The result determined by atomic absorption spectro-photometric method corresponded to roentgenographic and histopathological observations. Results suggest that during the process of resorption of BMG and DBM,bone morphogenetic protein(BMP) was released by diffussion,which induced mesenchymal cells originating from the dura meter to differentiate into chondrocytes and oste-oblasts, resulting in formation of new bone. The induced osteogenesis is considered to be a complex process regulated not only by BMP but also by various systemic and local growth factors.
