青藤碱对肾移植大鼠静脉血肿瘤坏死因子-α的影响

摘    要:目的:通过检测青藤碱(alkaloidsinomenine,SIN)对肾移植受体鼠静脉血中肿瘤坏死因子-α(TNF-α)表达水平的影响,探讨SIN对大鼠肾移植术后急性排斥反应的抑制作用及效应以及与环孢菌素A(cyclosprinA,CsA)是否存在协同效应。方法:建立近交系大鼠F344→Wistar模式的肾移植受体实验动物模型48只,术后按施加的处理因素不同将48只肾移植急性排斥反应动物模型分为4组[生理盐水(NS)组,SIN组,CsA组和SIN+CsA组],应用ELISA法检测静脉血TNF-alpha的表达水平;完整留取移植肾行病理切片,观察病理改变。结果:TNF-alpha的表达水平在SIN组低于NS组,差异存在显著性(P<0.05);SIN组与CsA组差异无显著性(P=0.530>0.05);SIN+CsA组低于SIN组及CsA组,差异存在显著性(P<0.05)。病理切片按移植肾排斥反应组织学分级标准,分级结果为,NS组3~4级;SIN组1~2级;CsA组1~2级;SIN+CsA组0~1级。结论:SIN对同种异体大鼠肾移植的急性排斥反应起到较为确切的免疫抑制作用,能明显下调TNF-alpha的表达水平,...

Abstract:Objective To observe the immunosuppressive effects of alkaloid sinomenine and its synergistic effects in combination with cyclosprin A in acute rejection after rat renal allograft via monitoring the levels of TNF-alpha in renal allograft rats. Methods The acute rejection models were established with inbored F344 being donors and inbored Wistar being recipients, 48 recipients were randomly divided into 4 groups. ELISA was used to test the levels of TNF-alpha quantitatively. The transplanted kidneys were used to observe the pathological changes. Results The levels of TNF-alpha was increased significantly in Group NS, and was higher than those in other two groups(P< 0.05). There was also significant difference between Group SIN and Group SIN CsA. Conclusion It is suggested that SIN has a significantly inhibitory effects and its better synergistic effects in combination with cyclosprin A in acute rejection after rat renal allograft.
