摘    要:[目的]研究人C2H2型锌指蛋白基因ZNF580在人脐静脉内皮细胞增殖和迁移中的作用.[方法]利用细胞转染技术获得瞬时过表达ZNF580的EA.hy926内皮细胞,并用半定量RT-PCR、Western blotting法检测ZNF580的表达情况;采用MTT比色法和细胞迁移实验检测ZNF580对内皮细胞功能的影响.[结果]获得瞬时过表达ZNF580的内皮细胞(转染效率为60%),且ZNF580基因过表达内皮细胞增殖和迁移能力分别增强3倍和2.5倍.[结论]ZNF580能够促进内皮细胞增殖和迁移.

Abstract:[Objective]To investigate the role of the novel human C2H2-zinc finger gene ZNF580 in the proliferation and migration of human umbilical vein endothelial cells(HUVEC). [Methods] By the induction of liposome, the plasmids pEGFP-ZNF580 was transfected into EAhy926 cells with Lipofeetamine TM 2000 for 48 h. The localization was estimated by fluorescence microscope. The expression of ZNF580 was evaluated by RT-PCR and Western blotting. MTT eolorimetric assay and cell migration assay were designed to investigate the effects of ZNF580 on endothelial cell growth and motility. [ Results]The EA.hy926 cells were successfully transfected with plasmids pEGFP-ZNF580 transiently(the transfeetion efficiency rate was 60%). ZNF580 enhanced endothelial cell proliferation and migration by 3 fold and 2.5 fold, respeetly. [ Conclusion ] These results elucidate the important role that ZNF580 may promote the migration and proliferation of endothelial cells, which provides a foundation for a novel approach to regulate angiogenesis.
