摘    要:
[目的]观察siRNA沉默VDAC1基因对乳腺癌MCF-7细胞增殖及细胞周期的影响.[方法]针对人VDAC1基因设计并合成siRNA,并将VDAC1-siRNA转染MCF-7细胞后,应用实时荧光定量PCR和Western印迹法检测MCF-7细胞中VDAC1的表达水平,MTT法检测MCF-7细胞的增殖情况,流式细胞术检测MCF-7细胞的周期分布,同时Western印迹法检测周期蛋白D1的表达.[结果]MCF-7细胞转染VDAC1-siRNA后能够有效抑制VDAC1的表达,显著抑制其增殖水平,促使MCF-7细胞发生G1期阻滞,同时下调Cyclin D1蛋白表达.[结论]VDAC1沉默能够有效抑制人乳腺癌MCF-7细胞增殖,促使细胞发生G1期阻滞.VDAC1可作为乳腺癌治疗的新靶点.

Abstract:
[Objective]To observe the effect of siRNA silencing VDAC1 gene on the proliferation and cell cycle of breast cancer MCF-7 cells.[Methods]siRNA was designed and synthesized for human VDAC1 gene and MCF-7 cells were transfected through VDAC1-siRNA.Real time fluorescent quantitative PCR and West-ern blot were used to detect the expression level of VDAC1 in MCF-7 cells;MTT assay was used to detect the proliferation of MCF-7 cells,the cell cycle distribution of MCF-7 cells was detected by flow cytometry,and the expression of cyclin D1 was detected by Western blotting [Results]VDAC1-siRNA transfected from MCF-7 cells can effectively inhibit VDAC1 expression,inhibit the proliferation level,promote MCF-7 cells to occur G1phase arrest and down regulate the expression of Cyclin D1 protein.[Conclusion]VDAC1 silencing can effec-tively inhibit the proliferation of human breast cancer MCF-7 cells,and promote cell arrest in G1phase,which can be used as a new target for the treatment of breast cancer.
