不同侵袭潜能卵巢癌细胞系蛋白质谱的比较

摘    要:目的探讨与卵巢癌侵袭潜能相关的蛋白质。方法将卵巢癌细胞系SKOV-3在裸鼠体内多次传代,体外培养建系,结合体外黏附、侵袭、移动实验,筛选不同侵袭潜能的细胞亚系,用表面增强激光解吸电离-飞行时间-质谱技术检测其蛋白质谱。结果SKOV-3F3细胞亚系较其母系SKOV-3侵袭潜能更高。有6个差异蛋白质峰(质荷比M/Z分别为6249、14675、15425、15717、17123和17660)仅在SKOV-3中表达,3个差异蛋白质峰(M/Z分别为4355、4823和5763)仅在SKOV-3F3中表达,2个差异蛋白质峰(M/Z分别为1842和3145)可在SKOV-3的3代亚系中捕获。此外,5个差异蛋白质峰(M/Z分别为1242、5465、6899、6568和14027)和1组蛋白质谱峰簇在SKOV-3和其3代亚系中均可表达,但强度不同。结论体内传代结合体外实验可筛选出不同侵袭潜能的卵巢癌细胞,它们之间的差异蛋白与侵袭潜能密切相关。

Abstract:Objective To study the proteins related to invasive potential of ovarian cancer cell line.Methods The nude mice were intraperitoneally injected with the human ovarian cancer cell line(SKOV-3). The metastatic tumors were cultured in vitro for cell line establishment and transplantion.We evaluated the invasive potential of cell lines by adhesive test and matrigel invasion assay.Different cell lines with different invasive potential were tested for by surface enhanced laser desorption/ionization-time of flight-mass spectrometry(SELDI-TOF-MS)proteinchip array technology and compared their proteomic fingerprint patterns.Results SKOV-3F_3 showed more invasive capacity than SKOV-3 in adhesive test in vitro and Matrigel invasion assay and so on.By comparing their proteomic fingerprint patterns,we identified six distinct proteins(6249,14675,15425,15717,17123 and 17660 at M/Z value) only expressed in SKOV-3.To the contrary,three distinct proteins(4355,4823 and 5763 at M/Z value)only expressed in SKOV-3F_3.Furthermore,two distinct proteins(1842 and 3145 at M/Z value) were only be detected in SKOV-3F_1,SKOV-3F_2 and SKOV-3F_3.Besides,five distinct proteins(1242,5465,6899,6568 and 14027 at M/Z value) and a mass cluster expressed in SKOV-3 and its sublines,but their abundance were different.Conclusion The ovarian cancer cell lines with different invasive potential could be screened by in vivo passaging combined with test in vitro.These distinct proteins were closely correlated with high invasive potential.
