小鼠粒-单系祖细胞体外培养的扫描电镜观察

摘    要:用液体法培养了小鼠的GFU—GM,并对培养不同时间的集落细胞进行扫描电镜观察。CFU—GM在分化成熟过程中有如下特点:(1)粒系细胞集落细胞表面由粗糙变光滑,主要变化在其表面的微绒毛。(2)单系细胞集落在培养早期成团,细胞表面光滑,4天后变粗糙,形状由规则变成多形性,由大小均匀至大小不一;培养中期有伪足形成.(3)同一集落位于中心的细胞较周边细胞幼稚.同时还发现粒细胞集落基底多存在一个贴壁细胞.

Abstract:We cultured CFU-GM of C57BL/6j mice in liquid media and observed the changes of cell surface morphology in granulocyte and macrophage colonies during the process of maturation with scanning electron microscope. The characteristics of cell surface morphology of CFU-GM were as following: 1. The surface of cells in granulocyte colonies changed from rough to smooth. The main changes occurred in microvilli. 2. The surface of cells in macrophage colonies changed from smooth to rough. The shapes of the cells were spherical in two-day's culture, but became triangular or spindle-shaped following maturation. The pseudopodia of the cells were formed after four-day's culture. The more mature the cells, the more numerous the pseudopodia. 3. In the same colony, the cells located at the periphery were more mature than in the center. Furthermore, we often observed there was a flattened and large accessorial cell under the granulocyte colony.
