青藤碱对脑缺血再灌注损伤大鼠环氧化酶-2表达和血脑屏障通透性的影响

摘    要:
目的 探讨青藤碱( sinomenine ,Sin)对缺血再灌注损伤大鼠脑组织保护作用及对环氧化酶-2 (cyclooxygenase-2 ,COX-2) 表达和血脑屏障通透性的影响.方法 将大鼠随机分为假手术组、缺血再灌注组、Sin低剂量治疗组和Sin高剂量治疗组,线栓法建立局灶性脑缺血再灌注模型.Sin低(Sin30 mg/kg)、高剂量(60 mg/kg)治疗组于术前30 min分别给予大鼠腹腔注射.用免疫组化法观察缺血90 min再灌注24 h大鼠额顶部皮质COX-2表达及脑含水量和伊文斯蓝(EB) 含量变化,并进行2,3,5-三苯基氯化四氮唑(TTC)染色观察脑梗塞体积变化和神经症状评分.结果 Sin高、低剂量治疗组神经功能缺失评分较缺血再灌注组降低,高、低剂量组之间差异亦具有统计学意义(P<0.05).Sin高、低剂量治疗组脑梗塞体积较缺血再灌注组减小,高、低剂量组之间差异亦具有统计学意义(P<0.05).与假手术组比较,缺血再灌注组额顶部皮质COX-2表达明显增加;与缺血再灌注组比较,Sin高、低剂量治疗组COX-2表达均显著减少(P<0.05),高、低剂量组之间差异亦具有统计学意义(P<0.05).与假手术组比较,缺血再灌注组脑含水量和EB 含量明显升高;与缺血再灌注组比较,Sin高、低剂量治疗组脑含水量和EB含量明显降低(P<0.05),高、低剂量组之间差异亦具有统计学意义(P<0.05).结论 Sin对缺血再灌注脑损伤具有保护作用,其机制可能是通过抑制再灌注损伤后COX-2表达,减轻血脑屏障通透性.
