| 摘 要： | 目的探索依曲替酸治疗银屑病的机制。方法免疫组化法检测银屑病皮损和正常表皮维生素D受体(VDR)蛋白表达;Western印迹法半定量检测依曲替酸对HaCaT细胞VDR蛋白表达的影响。结果银屑病皮损VDR蛋白表达水平(33.41±2.22)明显高于正常皮肤(14.87±3.18),差异有统计学意义(P<0.01);不同浓度依曲替酸作用HaCaT细胞24h,48h和72h后细胞VDR蛋白表达均较阴性对照组降低,呈明显的时间和剂量依赖性。结论银屑病皮损VDR蛋白表达异常,依曲替酸可降低HaCaT细胞VDR蛋白表达。 |

| Abstract: | Objective To explore the antipsoriatic mechanism of acitretin.Methods The expression of VDR protein were observed both in psoriatic epidermis and normal skin by immunohistochemical method.VDR protein was half quantitatively analyzed by western blot assay after culturing HaCaT cells exposed to acitretin.Results Immunohistochemical results showed that expression level of VDR protein in psoriatic lesion(33.41±2.22) was higher than that in normal epidermis(14.87±3.18)(P<0.01).Level of VDR protein in HaCaT cells was decreased after treated by acitretin with a time and dose.Conclutions The expression of VDR protein in psoriatic lesion is abnormal.Acitretin can decrease expression of VDR protein in HaCaT cells. |
