摘    要:目的 :观察尼莫地平对体外培养成骨细胞的影响。方法 :在新生SD大鼠头颅骨第 2继代成骨细胞 (OB2 )培养液中分别加入不同浓度 (10 - 1~10 - 9g·L- 1)尼莫地平 ,分别观察OB2 的增殖功能(用波长 5 70nm处OD值表示 ) ,分化功能 用碱性磷酸酶 (ALP)活性表示 ]和矿化功能 (用每视野矿化结节数量表示 )。结果 :增殖功能OD值为 0 .12 1±s 0 .0 0 9~ 0 .41±s 0 .0 4(P <0 .0 5 ,P <0 .0 1) ;ALP活性为 (0 .0 92± 0 .0 0 8)U·mg- 1Pro(P <0 .0 5 ) ;每视野矿化结节数量为 (1.3± 0 .9)个 (P <0 .0 1)。结论 :尼莫地平不同浓度对OB2 的增殖、分化和矿化功能作用各异 ,当尼莫地平浓度为 10 - 6 g·L- 1时 ,对OB2 的增殖和分化功能具有刺激作用 ,对OB2 的矿化功能具有显著的抑制作用。

Abstract:AIM:To obseve the effects of nimodipine on osteoblasts in vitro . METHODS: The culture medium with different nimodipine concentrations(10 -1 ~10 -9 g·L -1 ) and the second generation osteoblasts(OB 2) from the skull of newborn SD rat were mixed for the observation respectively about the proliferation (OD value at wavelength 570 nm), the differentiation(ALP activity) and the mineralization (mineralized nodes/field of vision) of the OB 2. RESULTS: The proliferation function was 0.121± s 0.009 ~0.41± s 0.04 OD value( P < 0.05, P < 0.01); the ALP activity was(0.092±0.008)U·mg -1 proteins ( P <0.05) and the mineralized nodes was (1.3±0.9)( P <0.01). CONCLUSION: In comparison with control group, the proliferation and differation of the OB 2 are separately increased by 10 -6 g·L -1 nimodipine concentration, but the minearlization is significantly inhibited. The proliferation of the OB 2 is significantly when nimodipine concentration escalates to the 10 -2 g·L -1 , the proliferation is not effected when nimodipine concentration descends to the 10 -8 g·L -1 .
