川芎嗪大鼠肝脏/肾脏缺血再灌注损伤中P-选择素表达的影响

目的研究川芎嗪对肝脏/肾脏缺血再灌注损伤中对P-选择素、肝脏组织MPO和NF-κB活性表达的影响。方法建立大鼠肝脏、肾脏缺血再灌注损伤模型,全自动生化分析仪检测再灌注后肝脏、肾脏功能,光镜下观察肝脏、肾脏病理学变化,和以免疫组化法检测肝脏肾脏组织中P-选择素的表达情况,肝脏组织MPO和NF-κB活性表达的情况。结果川芎嗪处理后肝、肾功能不全程度明显减轻,且肝肾组织中P-选择素表达减少(P0.05),肝脏组织MPO和NF-κB活性表达均明显降低(P0.05)。结论在肝脏/肾脏缺血再灌注过程中P-选择素、肝脏组织MPO和NF-κB活性表达的增高,是通过介导中性细胞向肝、肾组织中浸润聚集而在肝脏/肾脏缺血再灌注损伤中发挥重要作用。川芎嗪可以减轻肝脏/肾脏缺血再灌注损伤程度,其机制可能是抑制肝肾组织中P-选择素、肝脏组织MPO和NF-κB活性的表达。

Effects of ligustrazine on P-selectin expression of rats with liver/kidney ischemia-reperfusion injury

Objective To study the effect of ligustrazine on the expression or activity of P-selectin,myeloperoxidase(MPO) and NF-κB in liver/kidney tissue with ischemia-reperfusion injury.Methods Ischemic liver/renal injury in rats was made.The liver and renal function was measured with Automatic Chemistry Analyzer.Liver and kidney histology were observed by light microscope.The expression of P-selectin,MPO and NF-κB in Liver or kidney tissue were evaluated by immunohistochemistry.Results The liver and renal dysfunction were significantly lightened with ligustrazine pretreatment.Moreover,administration of ligustrazine significantly reduced the expression of P-selectin in liver /kidney(P<0.05),as well as significantly reduced the activity of MPO and NF-κB in liver(P<0.05).Conclusions During ischemia-reperfusion injury,the increasing of the expression or activity of P-selectin,MPO and NF-κB may play an important effect through the increased neutrophil aggregation in the liver and kindney.Ligustrazine can attenuate liver and kidney ischemia-reperfusion injury,and protective mechanism may partly inhibit the expression of P-selectin activity in liver/kidney tissue.
