甲壳胺提高难溶性药物溶出速率的研究

摘    要:本文以甲壳胺为辅料,采用混合研磨法,考察甲壳胺的粘度、用量、与药物混合研磨时间等因素对微粉化灰黄霉素溶出速率的影响。结果表明,灰黄霉素的溶出速率随甲壳胺粘度减小、用量增大、与药物混合研磨时间延长而增大。并且与淀粉作比较,效果优于淀粉。由X-衍射分析、差热分析可知,经混合研磨后灰黄霉素的晶型没有发生改变。此外,将甲壳胺分别与尼莫地平、吲哚美辛共同研磨,都可使它们的溶出速率明显提高。

Abstract:We studied the effect of chitosan on the dissolution rate of some insoluble drugs.We investigated the effects of viscosity,the chitosan-drug ratio(W/W),and the grinding time for the dissolution of micronized griseofulvin.The results were an increased dissolution rate of griseofulvin with a decrease of viscosity of chitosan,and an increase of the chitosan-drug ratio and the grinding time.The increased dissolution rate by chitosan was greater than that by starch.The X-ray diffraction patterns and DTA suggested no change in crystal form.Moreover,we studied the dissolution rate of nimodipine and indomethacin ground with chitosan respectively.They also showed an increased dissolution.
