摘    要:
目的 探讨内皮素(ET)、一氧化氮(NO)在动脉硬化闭塞症(ASO)形成和发展听 作用及ET、一氧化氮合酶(NOS)基因表达同ASO的关系。方法 收集40例ASO病人、120处病变血管和30例正常血管,根据病理学检查分为ASO早期、中期、晚期和正常血管4组。利用放射免疫测定法和Grisse方法检测ASO病脸的血浆ET和NO水平,利用逆转录-聚合酶链反应(RT-PCR)技术检测4组血管ETmRNAT

Abstract:
Objective To investigate the role of plasma endothelins (ET), nitric oxide (NO) in the pathogenesis of atherosclerosis (AS) obliterans (ASO) and the relationship between ASO and ET, NO synthase (NOS) gene expression. Methods 120 samples from 40 patients with ASO were collected and 30 samples of normal arteries from traumatic amputation and fresh cadavers. All the samples were divided into early, middle and late stage ASO group and normal control group according to pathology. The plasma ET and NO levels were determined by radioimmunoassay and Griss method in the ASO patients. The ET mRNA and NOS mRNA gene expression in the four groups was detected by RT PCR method. Results The ASO patients showed high level of plasma ET and low level of plasma NO. The ASO artery NOS enzyme activity and NOS mRNA expression were decreased significantly, while the ET mRNA gene expression increased obviously, all changes were found most obviously in the advanced ASO arteries.Conclusion The high ET expression and low NOS mRNA expression existed during the development of ASO. High ET level and low NO level occurred in the process of ASO pathogenesis.
