异氟醚后处理对盲肠结扎穿孔大鼠急性肺损伤的保护作用

摘    要:目的观察异氟醚(isoflurane,ISO)后处理对盲肠结扎穿孔术(cecal ligation and puncture,CLP)后大鼠急性肺损伤(acute lung injury,ALI)的保护作用。方法32只SD雄性大鼠随机分为4组(每组n=8):正常组(N组),未行任何处理;假手术组(Sham组),单纯行开腹关腹术;盲肠结扎穿孔术组(CLP组),行CLP建立大鼠ALI模型;异氟醚组(ISO组),建立CLP模型后4 h腹腔注射液态异氟醚2 ml/kg。术后12 h各组动物腹主动脉放血处死,右肺进行病理切片和肺湿/干比(W/D)测定,组织匀浆测超氧化物歧化酶(SOD)、丙二醛(MDA)和髓过氧化物酶(MPO)的含量;左肺行支气管肺泡灌洗,取上清液测总蛋白、肿瘤坏死因子α(TNF-α)和白细胞介素10(IL-10)的含量。结果肺组织病理学检测结果显示CLP组大鼠肺组织损伤严重,ISO组肺组织损伤较轻。与N组比较,CLP组和ISO组中SOD含量降低(P〈0.01),其余检测指标均增加(P〈0.01)。与CLP组比较,ISO组SOD和IL-10含量较高(P〈0.01);其余检测指标均低于CLP组(P〈0.05或P〈0.01)。结论在本实验条件下,大鼠腹腔注射液态ISO可发挥抗炎、抗氧化作用,对CLP所致大鼠ALI有部分保护作用。
