白芍多糖的制备、理化性质及抗肿瘤活性研究

摘    要:目的:研究中药白芍中多糖的制备、理化性质及对小鼠路易斯肺癌和S180肉瘤的抗肿瘤活性。方法:采用水提醇沉方法提取白芍中多糖(BSEP),测定其糖和糖醛酸含量、元素组成、单糖组成;同时采用小鼠路易斯肺癌足趾皮下接种模型和S180肉瘤腋皮下接种模型考察BSEP的抗肿瘤活性。结果:BSEP的提取率分别为1.56%,糖含量为80.56%,糖醛酸含量为3.33%,单糖组成分析为91.7%的葡萄糖,4.19%的半乳糖以及2.83%的阿拉伯糖。生物活性表明BSEP样品分别以200,100mg·kg^-1剂量ip×10qd的治疗方案,对小鼠体内路易斯肺癌足趾接种模型的肿瘤抑制率分别为54.58%和43.33%,对小鼠S180肉瘤皮下接种模型的肿瘤抑制率分别为47.08%和38.98%。结论:BSEP是主要以葡萄糖组成,且含有少量糖醛酸的多糖,具有一定的抗肿瘤活性,对小鼠路易斯肺癌的疗效稍优于S180肉瘤模型。

Abstract:Objective: To study the preparation, physicochemical properties and antitumor activity of polysaccharide from the root of Paeonia lactiflora Pall. Methods: Polysaccharide extracted from the root of Paeonia lactiflora Pall. with water precipitated by ethanol, and named BSEP. Its physicochemical properties, including the content of carbohydrate, elemental composition and monosaccharide composition, and antitumor activity were determined. Results: Extraction yield, sugar and uronic acid content of BSEP were 1.56% ,80. 56% and 3.33%, respectively. Glucose was the main monosaccharide. The bioactive assay showed that the inhibition rates of Lewis pulmonary carcinoma were 54. 58% and 43.33% ,and the inhibition rates of Sarcoma 180 were 47.08% and 38.98% when two doses of BSEP(200 and 100 mg. kg1) were administered intraperitoneadly (ip)every day for 10 d after the tumor inoculation, respectively. Conclusion: BSEP was a crude polysaccharide with much glucose and minor uronic acid. Antitumor activity of BSEP on Lewis pulmonary carcinoma was better than on Sarcoma 180 slightly.
