丹酚酸B与三七总皂苷配伍对缺氧复氧内皮细胞粘附分子-1表达及对中性粒细胞与内皮细胞粘附率的影响研究

摘    要:目的研究丹酚酸B与三七总皂苷配伍对缺氧复氧内皮细胞粘附分子-1(ICAM-1)表达及对中性粒细胞与内皮细胞粘附率的影响。方法体外建立大鼠心脏微血管内皮细胞缺氧再给氧模型,采用流式细胞仪(FCM)检测内皮细胞ICAM-1阳性细胞百分比及平均荧光强度;龙胆紫染色法检测中性粒细胞与血管内皮细胞粘附率。结果内皮细胞缺氧复氧后ICAM-1阳性细胞比例显著增加,ICAM-1平均荧光强度显著升高;150、450μg/L剂量组显著抑制ICAM-1表达和中性粒细胞与内皮细胞粘附率。结论丹酚酸B与三七总皂苷配伍对缺氧复氧内皮细胞ICAM-1表达具有抑制作用,可降低中性粒细胞与内皮细胞粘附率,对内皮细胞损伤具有一定的保护作用。

Abstract:OBJECTIVE: To study the effects of salvianolic acid B (SalB) and PNS (Panax Notoginseng Saponins) concomitantly on the expression of ICAM-1 of hypoxia/reoxgenation simulated endothelial cells (EC) and on the adhesion between PMN and endothelial cells. METHODS: The hypoxia/reoxgenation model of rats' cardiac microvascular endothelial cells was established in vitro. ICAM-1 positive cells' percentage and the mean fluorescent intensity were determined by flow cytometry (FCM); and the adhesion between PMN and endothelial cells was detected by gentian violent pigmentation. RESULTS: The positive cell expression percentages of endothelial cells after hypoxia/reoxgenation increased markedly and the mean fluorescent intensity of ICAM-1 elevated significantly. The expression of ICAM-1 and adhesion between PNM and EC were markedly inhibited at doses of 150μg/L and 450μg/L. CONCLUSION: The combination of SalB and PNS could inhibit the expression of ICAM-1 in endothelial cells after hypoxia/reoxgenation and decrease the adhesion between PNM and endothelial cell. Furthermore, it is protective to endothelial cells from damage.
