| 摘 要： | 目的：旨在获得人内皮抑素(endostatin)分泌型基因片段。方法：合成含信号肽的上游引物，用聚合酶链反应技术克隆人分泌型内皮抑素基因，10g／L琼脂糖凝胶电泳后回收，将其重组入PGEM-T载体，双脱氧末端终止法测定其核苷酸序列。结果：经Genbank分析证实，所获得的645bp基因片段序列属于信号肽及人内皮抑素功能区段基因，翻译为蛋白质序列正确。结论：本研究为应用内皮抑素基因进行肿瘤的抗血管生成治疗奠定了基础。 |

| Abstract: | Objective: To Procure gene fragment of secretive human endostatin. Methods: Synthesize the upper primer which contains the signal sequence, the gene of secretive human endostatin was cloned by PCR. After being identified by 10 g/L agarose gel electrophoresis, it was cloned into PGEM-T vector and sequenced according to dye primer sequencing kit. Results: Correct gene fragment of 645 bp was obtained and identified as functional section of signal and human endostatin gene by sequencing and Genbank analysis, the protein sequence was also correct. Conclusion: Experiment lays fundation for tumor antiangiogenesis therapy with endostatin gene. |
